MOLECULAR CHARACTERIZATION OF TOMATO (SOLANUM LYCOPERSICUM L.) GENOTYPES USING THE LECO014 SSR MARKER FOR DROUGHT TOLERANCE ASSESSMENT

Drought stress is one of the major abiotic factors limiting tomato (Solanum lycopersicum L.) production in many agricultural regions worldwide. The identification of drought-tolerant genetic resources is therefore an essential objective in modern tomato breeding. Molecular marker technologies, particularly simple sequence repeat (SSR) markers, provide reliable tools for evaluating genetic variation and supporting marker- assisted selection because they are highly reproducible, co-dominant, and unaffected by environmental conditions. In the present study, forty-nine tomato genotypes were characterized using the LEco014 SSR marker. Genomic DNA was extracted from young leaf tissues using the cetyltrimethylammonium bromide (CTAB) method, followed by polymerase chain reaction (PCR) amplification. The amplified DNA fragments were separated by 2% agarose gel electrophoresis and visualized under ultraviolet illumination. The PCR profiles confirmed the successful amplification of the target locus in most of the analyzed genotypes, demonstrating the applicability of the LEco014 marker for preliminary molecular characterization of tomato germplasm. The obtained results indicate that SSR marker analysis is a practical approach for assessing genetic variation and can support the selection of promising parental materials for drought-tolerance breeding programs. The integration of molecular marker information with physiological and agronomic evaluations is expected to improve the efficiency and accuracy of tomato improvement under water-limited conditions.
