G894T NOS III GENE POLYMORPHISM AND RESISTANT HYPERTENSION IN THE UZBEK POPULATION

The aim: to study the G894T gene polymorphism of the NOS3 gene on the risk of developing resistant arterial hypertension (AH) in Uzbek population. Materials and Methods: The study included 176 patients with I-III degree of AH, who are on outpatient treatment at the Republican Specialized Scientific and Practical Medical Center of Cardiology. Genomic DNA was isolated from whole blood using the DiatomTM DNA Prep 200 kit (Laboratory IsoGen, Russia) according to the manufacturer's standard protocol. The quantity and quality of isolated DNA were assessed using gel electrophoresis and a NanoDrop spectrophotometer (Thermo Scientific, USA). Gene polymorphisms were identified by real-time polymerase chain reaction using allele and gene-specific primers and probes.Results: An analysis using genetic models of inheritance revealed an association of the C allele T704C of the AGT gene polymorphism with resistant hypertension and a protective effect of the G allele G894T of the NOS3 gene polymorphism on the risk of developing resistant hypertension in the Uzbek population.Conclusion: C allele and CC genotype T704C of the AGT gene polymorphism are associated with resistant hypertension. Carrying the G allele and GG genotype G894T of the NOS3 gene polymorphism is associated with a low risk of developing resistant hypertension in Uzbek population.
