IN VITRO ANTISTAPHYLOCOCCAL EFFICACY OF A VANCOMYCIN-IMPREGNATED GELATIN SPONGE: ZONE OF INHIBITION AND 14-DAY RELEASE PROFILE

This microbiological in vitro study evaluated a novel vancomycin-impregnated absorbable gelatin hemostatic sponge designed for intraoperative local antibiotic delivery. Antimicrobial efficacy was tested against MSSA and MRSA using (1) a disc diffusion assay comparing the sponge to a standard 30 µg vancomycin disc and (2) a 14-day sustained-release model with daily eluate replacement and agar well diffusion testing. The sponge produced significantly larger inhibition zones than standard vancomycin discs at baseline and maintained measurable antibacterial activity throughout the full 14-day period, corresponding to progressive sponge degradation. These findings indicate that the gelatin sponge can achieve strong initial antistaphylococcal activity and prolonged vancomycin release, supporting its potential as a cost-effective local antimicrobial carrier for preventing or adjunctively treating surgical site infections in orthopaedic and other surgical fields.
