A study was conducted to investigate the effect of pre-sowing low temperature and light treatment (vernalization) on the phenological development and yield of winter wheat (Triticum aestivum L.). Seeds were vernalized at 4 °C for 4 weeks under light (VL) or dark (VD) conditions, followed by cultivation under Speed Breeding (SB) conditions. Seeds treated with VL exhibited faster root development and significantly accelerated vegetative growth. Flowering occurred 6 days earlier under VL compared to VD and control, and grain maturation was observed 13 days sooner. Phenotypic analysis showed that VL treatment increased seed dry weight and the number of effective panicles per plant by 28.16% and 20.23%, respectively, while overall yield increased by 21.87%.Transcriptomic analysis revealed that differentially expressed genes (DEGs) involved in gibberellin (GA) synthesis and signaling, as well as MADS-box transcription factors VRN-A1 and MADS55, played a key role in early flowering under VL treatment. These results confirm that the combination of low temperature and light during vernalization effectively accelerates vegetative growth, flowering, and yield of winter wheat, providing a foundation for the development of speed vernalization-based breeding technologies.
